Use a mailing list, not an alias.
On the list main tab, set list to only allow moderator to post 
Add other names to allowed posters ltab for the list

do I need to add all user for this under subscribers inorder to receive mail sending from poster list or moderator?
for example: allstaff@example.com is group mail and few user only have right to send mail using allstaff@example.com that i need to put in allowed posters tab that you highlight and for remaining usermailbox of domain to recieve mail from allstaff do i need to add in subscribers?

Yes, you need an API script (or difficult manual process) to keep the subscriber list populated with all active users.   

You will probably have some accounts that do not need to be included in "all", which will become clear over time.   Those accounts can be tagged with specific attributes, and then the script modified to look for those attributes when decided what to include in "all".  

 Accounts which cannot receive mail will be automatically dropped from the list when the bounce limit has been reached.   The drop even event can trigger an email to you as administrator, which you may find useful or may find overwhelming.

You can't disable the unsubscribe function. The best workaround we found was to create a "Mailing List Unsubscribe" event so at least the domain admins know when it occurs and can re-add the subscriber.

For an all-employees list, you probably want an API script that adds new hires.  It can pick up the unsubscribes in the same pass.
